چکیده | ||
بهمنظور تعیین ژنوتیپ متحمل و حساس به خشکی لوبیا در مرحله جوانهزنی، آزمایشی بهصورت فاکتوریل دو عاملی در قالب طرح کاملا تصادفی با سه تکرار در سال 1394 اجرا شد. عامل اول شامل ژنوتیپ (21 ژنوتیپ)، عامل دوم تنش خشکی (با استفاده از پلیاتیلنگلیکول 6000 در 2 سطح صفر و 148/0- مگاپاسکال) بودند. نتایج تجزیه واریانس برای صفات مورد بررسی (شامل طول ریشهچه، طول ساقهچه، وزن تر ریشهچه، وزن خشک ریشهچه، وزن تر ساقهچه، وزن خشک ساقهچه، سرعت رشد ساقهچه، سرعت رشد ریشهچه، نسبت طول ریشهچه به ساقهچه، اختلاف وزن خشک و تر ساقهچه و اختلاف وزن خشک و تر ریشهچه) نشان داد که اثر ژنوتیپ و همچنین اثر متقابل ژنوتیپ و خشکی، بر کلیه صفات مورد بررسی معنیدار بودند. بر اساس نتایج حاصل از تجزیه خوشهای بهروش Ward، ژنوتیپهای مطالعهشده تحت شرایط نرمال و تنش خشکی از نظر خصوصیات گیاهچهای در چهار گروه مختلف قرار گرفتند که ژنوتیپهای غلاف سیاه و پاک بهترتیب بهعنوان ژنوتیپهای متحمل و حساس در شرایط تنش خشکی انتخاب شدند. بررسی الگوی بیان سه ژن GR، PRX و APX واکنشدهنده به تنش خشکی در دو ژنوتیپ پاک و غلاف سیاه لوبیا با استفاده از تکنیک Real time PCR مورد مطالعه قرار گرفت. پس از استخراجRNA و سنتز cDNA، تظاهر بیان سه ژن در تیمارهای صفر، 72، 144، 192 و 240 ساعت خشکی نشان داد که ژنهای واکنشدهنده به خشکی در این پژوهش بیان بالاتری در رقم متحمل نسبت به رقم حساس داشت و میزان بیان سه ژن PRX، GR و APX در ساعت انتهای تنش، در ژنوتیپ متحمل بهترتیب دو، چهار و شش برابر بیشتر از ژنوتیپ حساس بود. | ||
