ارزیابی تیمارهای مختلف هورمونی و سرمایی بر برخی شاخصهای بیوشیمیایی و جوانهزنی بذور سه گونه درمنه یکساله (Artemisia annua)، دوساله (A.biensis willd.) و چندساله (A.vulgaris L) | ||

چکیده | ||
درمنه گیاهی ارزشمند دارویی و مرتعی از تیره کاسنی دارای مصارف متعدد دارویی و غذایی است. باتوجه به وجود خواب در بذر این گیاه، پژوهش حاضر بهمنظور بررسی اثرات پرایمینگهای سرمادهی مرطوب و هورمونی روی شاخصهای بیوشیمیایی و جوانهزنی بذور سه گونه یکساله (annua)، دوساله (biensis) و چندساله (vulgaris)، انجام شد. این آزمایش به صورت کرتهای یکبار خردشده در قالب طرح پایه کاملاً تصادفی با سه تکرار در آزمایشگاه مرکز تحقیقات گیاهان دارویی و معطر دانشگاه آزاد اسلامی واحد شهرکرد در سال 1403 و بطور دو بار مجزا انجام شد. سطوح سرمادهی مرطوب در کرتهای اصلی و تیمارهای هورمونی شکستن خواب در کرتهای فرعی قرار گرفتند. سطوح سرمادهی مرطوب شامل عدم سرمادهی، 10، 20 و 30 روز سرمادهی بود. تیمارهای شکست خواب شامل هورمونهای اسید جیبرلیک (GA3) (120 میلی گرم در لیتر)، بنزیل آدنین (BA) (4-10مولار)، کینتین (KIN) (4-10مولار)، GA3 (120 میلیگرم در لیتر)+ بنزیلآدنین (4-10مولار)، GA3 (120میلیگرم در لیتر)+KIN (4-10مولار)، BA (4-10مولار)+KIN (4-10 مولار)، GA3 (120 میلیگرم در لیتر)+BA (4-10مولار)+KIN (4-10مولار) و شاهد (آب مقطر)، بودند. صفات مورد اندازه گیری در این آزمایش شامل شاخصهای بیوشیمیایی مقادیر آنزیمهای آلفا آمیلاز، بتا-1 و 3-گلوکوناز و پراکسی هیدروژن و شاخصهای جوانهزنی از جمله طول و وزن خشک گیاهچه، درصد جوانه زنی، بنیه بذر، میانگین مدت جوانهزنی، شاخص جوانه زنی بودند. نتایج نشان داد که سرمادهی مرطوب به طور معنیداری باعث افزایش صفات مورد ارزیابی و کاهش متوسط زمان لازم برای جوانهزنی شد. سرمادهی مرطوب به طور معنیداری باعث افزایش صفات مورد ارزیابی و کاهش متوسط زمان لازم برای جوانهزنی شد. بیشترین و کمترین مقادیر صفات مهم جوانهزنی از جمله طول گیاهچه (12/11-19/4 سانتیمتر)، شاخص بنیه بذر (9/7-46/1)، وزن خشک گیاهچه (14/101-88/39 میلیگرم)، جوانهزنی (12/71-63/29 درصد)، شاخص جوانهزنی (66/18-25/3)، مقادیر آلفا آمیلاز (77/11-41/2 میلیمول بر گرم وزن تر بذر)، بتا-1 و 3-گلوکوناز (81/9-57/2 میلیمول بر گرم وزن تر بذر) و پراکسید هیدروژن (63/0-191/0 میلیمول بر گرم وزن تر بذر) به ترتیب در تیمار تلفیقی سرمادهی مرطوب (20 و 30 روز)+تیمار هورمونی BA+KIN+ GA3 و تیمار شاهد بوجود آمد که نسبت به شاهد، افزایشی بیش از 100 درصد را نشان داد. مشخص شد که در غالب صفات مورد ارزیابی گونه یکساله بهتر از گونه دو ساله بهتر از گونه چندساله بود. بیشترین میزان جوانهزنی در گونه یکساله و کمترین آن در گونه چندساله مشاهده شد. بر اساس اثر متقابل سرمادهی و تیمارهای شکست خواب، 20 روز سرمادهی مرطوب و عدم تنش خشکی با کاربرد هر یک از تیمارهای BA، KIN و GA3 جهت جوانهزنی بذور درمنه و ایجاد بالاترین مقادیر شاخصهای بیوشیمیایی، مناسب به نظر میرسد. | ||
