چکیده | ||
Vibrio cholerae یک باکتری بیماریزای آبزی است که سویههای مختلف آن باعث ایجاد بیماریهایی چون وبا، اسهال حاد، مننژیت و سپتیسمی میشوند. با توجه به مقاومت سویهها به آنتی بیوتیکهای گوناگون، تولید واکسنهای موثر بر تمامی سویههای بیماریزا میتواند یکی از بهترین روشهای مبارزه باشد. از این رو، در این مطالعه پتانسیل واکسن بودن پروتئین VceC علیه باکتری V. cholerae با استفاده از روشهای بیوانفورماتیکی مورد بررسی قرار گرفت و در کنار تعیین ویژگیهای آنتیژنی، ویرولانسی و فیزیکوشیمیایی پروتئین نام برده، نقشهبرداری اپیتوپهای B و T به منظور طراحی بهترین واکسن زیرواحدی انجام شد. نتایج نشان داد که این پروتئین دارای ویژگیهای بهینه از جمله حضور در سطح تمامی سویههای مورد مطالعه، پایداری و نیمه عمر بالا، مقاومت به دما و همچنین حلال بودن در هنگام بیشبیانی است. همچنین نقاط آنتیژنیک و اپیتوپهای خطی و فضایی زیادی در طول توالی و ساختار پروتئین مشاهده شد. با توجه به موقعیت مکانی و ویژگیهای آنتیژنیک، دو واکسن زیرواحدی خطی و فضایی طراحی شد که میتوانند علیه بیماریزایی سویههای مختلف باکتری V. cholerae مورد آزمایش و استفاده قرار گیرند. | ||
