مقایسه تاثیر تکنیکهای مختلف پرایمینگ بر بهبود جوانهزنی و فعالیت آنزیمهای آنتیاکسیدانت در گیاهچههای گاوزبان اروپایی | ||

چکیده | ||
یکی از مشکلات تولید گیاهان دارویی از قبیل گاوزبان اروپایی، جوانهزنی غیریکنواخت و استقرار ضعیف گیاهچهها است. پرایمینگ یکی از راهکارها جهت بهبود جوانهزنی و رشد گیاهچهها در گیاهان زراعی و دارویی است. این پژوهش بهمنظور بررسی تاثیر پرایمینگ بر جوانهزنی، رشد و خصوصیات بیوشیمیایی و آنزیمی گیاهچههای گاوزبان اروپایی انجام گرفت. تیمارهای آزمایشی شامل هیدروپرایمینگ بهمدت 48 ساعت، ویتامینپرایمینگ (اسیدآسکوربیک 85/0 میلیمولار) بهمدت 48 ساعت، هورمونپرایمینگ (اسیدسالسیلیک 4 میلیمولار) بهمدت 60 ساعت، هالوپرایمینگ (نیترات پتاسیم 20 میلیمولار) بهمدت 24 ساعت و شاهد بودند. نتایج نشان داد که کلیه تکنیکهای پرایمینگ مورد استفاده، باعث افزایش معنیدار جوانهزنی، رشد، شاخصهای بیوشیمیایی و آنزیمی گیاهچهها گردید. همچنین استفاده از روشهای مختلف پرایمینگ در بذرهای گاوزبان اروپایی، موجب کاهش معنیدار میانگین زمانجوانهزنی و درصد گیاهچههای غیرنرمال گردید. بیشترین درصد و سرعت جوانهزنی، شاخص قدرت و طول گیاهچه از بذرهای هیدروپرایمشده بهدست آمد که بهطور معنیداری بیشتر از بذرهای شاهد بودند. همچنین، هیدروپرایمینگ موجب کاهش 80 درصدی گیاهچههای غیرنرمال و 25 درصدی میانگین زمانجوانهزنی گردید. فعالیت آنزیمهای کاتالاز، پراکسیداز و میزان اسیدآمینه پرولین در گیاهچههای حاصل از بذرهای پرایمشده بهطور معنیداری بیشتر از شاهد بود. بیشترین فعالیت آنزیم کاتالاز و محتوی پرولین بهترتیب در تیمار بذر با نیترات پتاسیم 20 میلیمولار و اسید سالسیلیک 4 میلیمولار مشاهده شد که اختلاف معنیداری با هیدروپرایمینگ نداشتند. هیدروپرایمینگ موجب افزایش 7/2 برابری فعالیت آنزیم پراکسیداز گیاهچهها گردید. بهطور کلی، در بین تیمارها، هیدروپرایمینگ بهمدت 48 ساعت را میتوان بهعنوان بهترین تیمار برای بهبود خصوصیات جوانهزنی، رشد، خصوصیات بیوشیمیایی و آنزیمی گاوزبان اروپایی در نظر گرفت. | ||
